Abstract: 

Background: We started with RNA-seq analysis and aimed to investigate the possibility of secretory protein matrix metalloproteinase-3(MMP3) as a new diagnosis and therapeutic target in cervical cancer. Methods: The study was conducted on Nov 2021 at the Second Affiliated Hospital of Qiqihar Medical University, Qiqihar, China. Through conjoint analysis of gene expression data as well as survival rate data, we explored the potential secretary proteins associated with cervical cancer carcinogenesis. One hundred patients aged 38-72 years with clinical stage I-IV cervical cancer, and 100 age-matched healthy women were included. The expression changes in serum of clinical patients was detected. We knockdown or overexpressed the secretory proteins then explored its influence on biological function of cervical cancer cells. Results: By cross-analysis of The Cancer Genome Atlas (TCGA) database and MetazSecKB database, MMP3 gene was most significantly upregulated in cervical cancer patients (P < 0.05). Furthermore, MMP3 protein was remarkably increased in the serum of clinical cervical cancer patients and decreased after receiving treatment. Overexpression of MMP3 in HT-3 cells or culturing new cells using the supernatant of the medium after MMP3 overexpression could increase cell viability (P < 0.05) as well as proliferation (P < 0.05). Knockdown of MMP3 reduced the phosphorylation of PI3K as well as AKT proteins, while the PI3K phosphorylation inhibitors could suppress the impact of MMP3 on increasing cell proliferation as well as viability. Conclusion: MMP3 could be an underlying target for early diagnosis and treat cervical cancer in the future.

Abstract: 

Colorectal cancer is the most prevalence malignancy that affect on stomachic system. After lung and breast cancer, colorectal cancer is the most common of death in the western world. Colorectal adenocarcinoma accounts for over 90% of the malignant tumors of the large bowel. Modification in cell adherent molecules and enzymes that degrade intra cellular membrane have important role for colorectal tumor creation. Matrix metalloproteinase enzymes degrade different component of extra cellular matrix and cause tumor creation, cancer development and metastasis. MMP3 overexpression due to single nucleotide polymorphism (SNP) 5A/6A at the -1171 position of the MMP3 gene promoter effects on tumor malignancy and cancer development. A case- control study was performed including 120 cancer patients from Emam Khomeini hospital in Tehran and Seid Alshohada hospital in Isfahan. After MMP3 promoter genotyping, the results were analyzed by SPSS 12.0 software. The colorectal cancer patients were divided in to two groups: M+non- metastasis patients and M-, the metastasis-free patients. The MMP3 genotypes were similar between controls and patients (P=0.43, OR=0.78, 95%CI, 0.37-1.64), but the 5A/5A genotype was more prevalent in the M+groups than controls (P=0.74, OR=2.91, 95%CI, 0.94-8.98). This frequency was more prevalent than M- (P=0.65, OR=1.90, 95%CI, 0.69-5.2). So we supposed the MMP3 5A/6A promoter polymorphism doesn' t appear to influence colorectal among colorectal cancer patients but 5A/5A genotype with MMP3 over-expression cause colorectal cancer development and metastasis.

Abstract: 

Introduction: Lichen planus is an inflammatory chronic illness with unknown cause that can irritate the oral mucosa. P53 is associated with malignant changes in oral epithelial cells. MMPs can destroy intercellular junctions and cause acantholysis. It seems that MMP-3 plays a significant role in this regard. The purpose of this study was to evaluate the salivary levels of P53 and MMP-3 in the patients with Oral Lichen planus (OLP) compared with the control group. Materials and Methods: 30 salivary samples were collected from patients with OLP (15 with erosive Oral Lichen planus (EOLP) and 15 with reticular Oral Lichen planus (ROLP)) and 30 salivary samples from healthy people as a control group. The salivary P53 and MMP-3 level was assayed by ELISA method. Statistical analysis of the Student’ s t-test, ANOVA and Pearson correlation coefficient was performed. Results: The salivary concentrations of P53 and MMP-3 in patients with EOLP were significantly higher than patients with ROLP and control group, but no significant difference was found between control group and patients with ROLP. Conclusion: The salivary concentrations of P53 and MMP-3 were significantly different between different clinical types of OLP.

Abstract: 

Background: A cell requires energy to do its work. Mitochondria are the energy-producing machines of the cell. Uncoupling proteins (UCPs) located in the inner mitochondrial membrane cause a proton leak from the intermembrane space to the matrix. Clearly, the changes in the proton gradient across the inner membrane affect ATP production. In this paper, the role of these proteins in different tissues as well as peripheral and central nervous tissues has been reviewed.Methods: This study has reviewed 63 publications explaining the various functions of UCPs in different tissues using PubMed, Elsevier, NCBI and EBSCO databases.Findings: Several studies have shown that UCPs, based on the type of tissue, change the cell activity.Conclusion: Studies related to UCPs roles in the nervous system are mostly restricted to the in-vitro situations; thus more investigation seems to be needed to reveal the UCPs actions in the in-vivo conditions of physiologic and pathologic studies. These studies potentially can open some new therapeutic strategies and hopes to cure neurodegenerative diseases.

Abstract: 

Background: Endometriosis is identified as presence of the endometrium outside the uterine cavity. Retrograde menstruation contributes to the endometrial tissue implantation and the establishment of endometriotic lesions at ectopic sites. It has been suggested that the endometriotic lesions are rich in angiogenic growth factors, while they have an essential role in survival and invasion of these cells. We investigated regulation of microRNA-93 (miR-93) and its involvement with vascular endothelial growth factor A (VEGFA) and matrix metalloproteinase (MMP) 3 expression in women with endometriosis. Materials and Methods: This was a cross-sectional study at Central Surgical Installation, Dr. Cipto Mangunkusumo General Hospital, Jakarta, Indonesia, between October 2020 and November 2021. Eutopic and ectopic endometrial tissues were collected from 30 subjects with laparoscopically-confirmed endometriotic women. Normal endometrial cells of non-endometriosis women served as controls. Total RNA was isolated from all samples and a quantitative reverse-transcription polymerase chain reaction (qRT-PCR) was used to analyze the expression of miR-93, VEGFA and MMP3. Results: There was no significant difference in the expression levels of VEGFA (2. 14 ±,0. 50, P=0. 719) and MMP3 (2. 99 ±,0. 42, P=0. 583) between endometriotic lesions of endometriosis women and the healthy endometrium. Expression of miR-93 was significantly lower in the eutopic endometrium (16. 7 fold) and ectopic endometriotic lesion (20 fold) compared to the normal endometrium (P<0. 001). Furthermore, we also observed a significant correlation between miR-93 and VEGFA expression in eutopic endometrium obtained from women with endometriosis (r=-0. 544, P=0. 029). Expression of the miR-93 was also negatively correlated with MMP3 expression in both eutopic (r=-0. 412, P=0. 01) and ectopic (r=-0. 539, P=0. 03) endometrial cells of women with endometriosis. Conclusion: VEGFA and MMP3 expression levels trended to be increased in both eutopic and ectopic endometrial tissues of endometriosis women, while down-regulation of miR-93 might be involved in the alteration of VEGFA and MMP3 in endometriosis.

Abstract: 

Bone gla protein and matrix gla protein are of common origin with evolutionary relation and have similar structural features and are functional in the formation of skeletal structures The present study have been evaluated the genes encoding vitamin k-dependant proteins (VKDPs) expression at hatching time (0), 1, 3, 6, 10, 12, 14, 20, 30 and 50 in skeletal Persian sturgeon sturgeon (Acipenser persicusBordin, 1897). The results of quantitative real-time PCR showed that the transcripts of VKDPs genes including bone Gla protein (bgp) and matrix Gla protein (mgp ) were significantly up-regulated during the transition to the exogenous feeding, confirming the hypothesis about the relevance of the above-mentioned genes in chondrogenesis at early developmental stages. Maximal levels of bgp and mgp expression at juvenile stage, indicating the importance of these genes in the mineralization of the skeleton in sturgeons, although these genes are also expressed in other body tissues. This information can be considered as a reference for other studies evaluating the quality of larvae and the influence of rearing biotic and abiotic factors in the skeletogenesis of this sturgeon species and the occurrence of skeletal deformities.

Abstract: 

Introduction: Reactive oxygen species (ROS) have been suggested to play an important role in the myocardial damage induced by ischemia-reperfusion. One element believed to be activated by ROS and to contribute to the reduction of ROS production, is the uncoupling protein-2 (UCP2). The objective of this investigation was to explore the effect of myocardial ischemia reperfusion on cardiac UCP2 mRNA and protein levels.Methods: Male Wistar rats (250-300gr) were subjected to 30 min occlusion and 2 hours reperfusion of left coronary artery. The expression of UCP2 mRNA and protein in the ischemic area of left ventricle and non ischemic area from right ventricle were analyzed. The mRNA and protein expression were determined by RT-PCR and western blotting, respectively.Results: Compared to control hearts, exposure to myocardial ischemia reperfusion caused a significant increase in UCP2 protein in the ischemic area of the left ventricle (116%±18, P<0.001), however UCP2 mRNA expression did not change significantly. Furthermore, in the non ischemic area of the right ventricle, neither protein nor mRNA levels were affected by myocardial ischemia reperfusion.Conclusion: We conclude that following acute myocardial ischemia reperfusion, UCP2 protein level is increased in the ischemic area of the left ventricle but not in the non ischemic area of the right ventricle, suggesting the local effect of ischemia on UCP2 protein expression. Furthermore, the discordance between mRNA and protein expression of UCP2 suggests that post transcriptional regulation of mRNA influences protein induction.

Abstract: 

Background & Aim: The patients suspected of appendicitis are evaluated by history, physical examinations and laboratory results. The aim of this study was to assess the diagnostic value of C-reactive protein in patients with suspected acute appendicitis.Patients & Method: In a prospective study, 299 patients with acute abdominal pain who had been admitted to hospital for observation were enrolled. Serum CRP measurements were performed and the results were compared with the final results of histopathology or follow-up results. The results were analyzed using Chi-square test and their accuracy, sensitivity, specificity, and positive and negative predictive values were determined.Results: The findings showed that the accuracy of CRP test in diagnosing appendicitis was 70% (CI=Confidence Interval 95% from 65% to 74.4%), the sensitivity was 80.7%(CI95% from 71.2% to 87.6%), the specificity was 65.4% (CI95% from 58.8% to 71.5%), and the positive and negative predictive values were 49.3% (CI95% from 41.3% to 57.4%) and 89.0%(CI95% from 83.2% to 93.0%) respectively. The odds ratio was 8 (CI95% from 4.3 to 14.4).Conclusion: The results of CRP test can help a surgeon rule out or confirm the diagnosis of appendicitis in suspicious cases.

Abstract: 

زمینه و هدف: پلی مورفیسم های Q/R192 آنزیم پارااکسوناز 1 (PON1) و 5A/6A آنزیم متالوپروتییناز ماتریکس (MMP3) 3 ممکن است با استعدادپذیری با بیماری عروق کرونری مرتبط باشند؛ جهت بررسی اهمیت این پلی مورفیسم ها در پاتوژنز بیماری عروق کرونری ما به مطالعه ارتباطی این پلی مورفیسم ها با CAD و تعداد رگ های بیمار در بیماران مبتلا به CAD پرداختیم.روش بررسی: پلی مورفیسم های ژن های PON1 و MMP3 را با روش PCR و Restriction Fragment Length Polymorphism (RFLP) در مبتلایان به CAD مطالعه کردیم. این پلی مورفیسم ها در 129 بیمار مبتلا به CAD و 115 نفر کنترل که تحت آنژیوگرافی عروق کرونری قرار گرفتند تعیین گردیدند. CAD به صورت وجود گرفتگی <%50 در حداقل یکی از عروق کرونری اصلی مشخص گردید و افراد با گرفتگی بیش از 10 درصد به عنوان کنترلعمل نمودند. یافته ها: فراوانی ژنوتیپ های PON1 192RR و MMP3 6A/6A در گروه بیماران مبتلا به CAD در مقایسه با افراد کنترل بیشتر بود (به ترتیب p<0.05 و (p<0.001 و ژنوتیپ ترکیبی RR/6A6A در افراد مبتلا به CAD از فراوانی بالایی در مقایسه با افرادی که نه ژنوتیپ MMP3 6A/6A و نه ژنوتیپ PON1 RR را داشتند برخودار بود (p<0.001) و در نهایت در مطالعه ارتباط ژنوتیپ ها با شدت بیماری، توزیع ژنوتیپ های PON1 192 و MMP3 5A/6A با تعداد رگ های بیمار مرتبط نشد (p>0.05).نتیجه گیری: پلی مورفیسم های ترکیبی PON1 192 و MMP3 5A/6A با بیماری CAD مرتبط می باشند اما تاثیری بر روی تعداد رگ های بیمار ندارند.
